Offer Description

Project title:Together or not? The relevance of the interplay of staphylocoagulase and staphylokinase in the development of Staphylococcus aureus infections.

Work in a project realized in collaboration between the two leading Polish universities: Jagiellonian University Krakow and University of Warsaw. The project (funded by the Opus grant of the National Science Centre) focuses on understanding virulence of Staphylococcus aureus, a leading pathogen responsible for serious human infections. In particular, the project focuses on mechanisms and outcomes of Staphylococcus taking control over the human coagulation (clotting) and fibrinolysis (clot dissolution).

Most important parts of the project are:

  • analysis of coagulation and fibrinolysis in a mouse model of infectious endocarditis caused by S. aureus (this will use transgenic mice expressing human plasminogen, allowing for staphylococcal proteins to fully activate fibrinolysis in these mice);
  • understanding the effects of coagulation and fibrinolysis during interaction of S. aureus with host immune system;
  • identification of the role of coagulation and fibrinolysis in formation of S. aureus biofilms.

The project will use an array of various techniques:

  • ex vivo (isolation and analysis of human leukocytes interacting with staphylococcal clots, analysis of tissues from human patients);

in vitro (bacterial protein purification, clotting measurements, laboratory models of biofilms and abscesses, confocal and fluorescent microscopy).

Skills/Qualifications

1. PhD Degree (or equivalent) in biology, microbiology, biotechnology, biochemistry or related fields. Alternatively, anticipated completion of PhD prior to work start date;

2. Experience working with laboratory mice and/or experience with (micro)surgery techniques and/or mouse models of heart and other cardiovascular diseases. Familiarity with basic laboratory techniques (e.g. FACS, ELISA, PCR, gene cloning), documented with results in prior publications;

3. An additional asset would be experience working with bacteria (especially welcomed: Staphylococcus aureus), and/or bacterial biofilms, and/or immune cells, and/or fluorescent and confocal microscopy;

3. Fluency in written and spoken English;

4. Reliability, good engagement, ability to independently plan, perform and document experimental work.

Specific Requirements

The post-doc will be responsible for:

  • analysis of the available literature, planning and performing experiments, and interpretation of their results;
  • present results at team meetings, seminars and conferences, and through scientific publications;
  • supervise students engaged in the project and coordinate project's work;
  • if possible – participate in additional trainings and research exchanges with partner institutions (University of Warsaw, Leuven University Belgium)

The project will be performed under guidance of dr. Jakub Kwiecinski (project leader) in collaboration with dr. Marta Zapotoczna (University of Warsaw).
